Abstract Teleost fish are organisms that generally show an indeterminate growth limit and are present in different biomes with different environmental conditions. Approximately up to 80% of the body composition of fish is formed by muscle tissue. Muscle tissue is composed of muscle fibers and undifferentiated satellite cells, forming a complex tissue that shows a widely varied shape depending on the species which form this group of animals. An important point to be considered is that the muscle growth of the fish is influenced by the environmental conditions in which they are inserted. This influence occurs at the molecular level, affecting the transcription and functionality of different transcripts related to myogenesis at different stages of fish development. This review aims to discuss some of the key aspects about the influence of environmental factors on the development and muscle growth of teleost fish.

RESUMO

Given the importance of the Golden kingklip (Genypterus blacodes) as an alternative to the production of salmonid species for worldwide diversification of aquaculture this study researched the morpho-histology of the digestive canal, providing a uniform nomenclature and basic information to support their nutritional management in captive conditions. The objective was to describe the normal morphology of the digestive canal. Twelve wild specimens were collected and dissected. Samples of the esophagus, stomach, liver, pyloric blind and intestines were obtained. An anatomical and histological description was performed. The digestive canal presented a short tubular esophagus, a slightly dilated, J-shaped stomach and thick walls. The stomach had a cardiac region that ended in a pyloric sphincter from which the intestine of thin walls. The intestine presented an anterior, middle and posterior segment. The anterior region of the first segment of the middle intestine presented 4-5 pairs of pyloric cecum. The middle intestine represented approximately 30 % of the total length of the digestive canal, with a smaller diameter compared to the first segment. The posterior intestine was separated from the middle intestine by a thick and powerful sphincter. The liver was large and elongated, and located in the peritoneal cavity located to the left plane and ventral to the swim bladder. The gallbladder was located lateral to the hepatic anterior wolf. The bile duct led to the duodenal region of the anterior intestine. Ventrally, the rectum ended in the anus, located in front of the anal fin. In general, the histological structures were variable according to the segment studied, a mucous tunic was recognized that presented a simple columnar epithelium, a lamina propria composed of loose connective tissue, a muscular layer of the mucosa composed of smooth muscle tissue and a submucosal layer composed of loose connective tissue, a circular and longitudinal stratum of muscular tunic, which had striated musculature and an adventitial tunic composed of a simple squamous epithelium. In conclusion, Golden kingklip (Genypterus blacodes) presents a digestive system typical of a carnivorous benthic fish adapted to feed on fish and crustaceans.

Introduction: Ornamental Angel fish (Pterophyllum scalare) is one of the main commercialized species from South America, with an important number of national and international dealers. Objective: To make a histological and morphometric description of P. scalare digestive tract. Methods: Nineteen adult individuals were used. Intestinal coefficient values were obtained, and a macroscopic and microscopic description of esophagus, stomach, and intestine (anterior, middle and posterior) was carried out. Samples were processed for paraffin embedding, and 5 µm sections were stained with H&E and P.A.S. Positive P.A.S. cells were counted. Lumen area and tissue area were measured in representative digitalized sections from each region. Results: P. scalare exhibited a short esophagus, followed by a blind bag shaped, scarcely muscular stomach, and a short intestine (intestinal coefficient 1.66 +/- 0.52). Four constitutive tissue layers were identified: mucosa, submucosa, muscular and in some areas also a serosa layer. Esophagus exhibit abundant globet P.A.S. + cells and shows the larger tisular area was observed. Luminal area and perimeter are larger in anterior intestinal region. Conclusions: The intestinal coefficient value (1.66 +/- 0.52), and the histological and morphometric characteristics of the evaluated organs, suggest that P. scalare is omnivorous with soft prey preferences.

RESUMO

Descriptive studies of the fish digestive system are fundamental because they provide information on the biology of the species. Thus, the objective of this study was to morphologically describe the digestive system of the pufferfish, Chilomycterus spinosus spinosus. For this, adult specimens of pufferfish (n = 10) of both sexes were used. The animals were fixed with 10% aqueous formaldehyde solution, dissected, analyzed descriptively and photographed. The results demonstrate that the pufferfish has a morphologically modified digestive system, which is adapted to the defense behavior. This species presents a pouch-shaped diverticulum, that is called abdominal pouch, which allows the expansion of the celomatic cavity and the temporary storage of food. Although it is used to store food, macroscopically the abdominal pouch does not show gastric folds. However, this absence is compensated by a small intestine containing innumerable villi.

RESUMO

The Gymnotus inaequilabiatus is a Neotropical fish widely distributed in marginal areas of bays. The aim of this study was to describe the main histological and histopathological traits in the head and exocrine kidney. Here, histometric and structural density techniques were associated with renosomatic index (RSI). The kidney was processed for light microscopy. Lipofuscin and hemosiderin content were visually estimated in the melonomacrophages centers (MMCs). All the biometric body variables were correlated with RSI, especially the kidney weight and gross lesions count. The general architecture of head and exocrine kidney was similar to that described for other teleost species. MMCs were prevalent in both portions and correlated with RSI in the head and exocrine kidney. Granulomatous structures were often observed in both portions; however, they were associated only in the exocrine kidney with RSI. Of all the structures hystometrically estimated, only proximal tubular diameter and thickness, and distal tubular thickness were correlated to renosomatic index. The RSI is an useful biometric variable that represent some physiological and morphological characteristics of kidney in G. inaequilabiatus. These findings may be used in future studies to evaluate the effects of environmental stressors on the renal adaptative physiological process.(AU)

Background: Peptidoglycan (PGN) recognition proteins (PGRPs) are important pattern recognition receptors of the host innate immune system that are involved in the immune defense against bacterial pathogens. PGRPs have been characterized in several fish species. The PGN-binding ability is important for the function of PGRPs. However, the PGRP-PGN interaction mechanism in fish remains unclear. In the present study, the 3-D model of a long PGRP of half-smooth tongue sole (Cynoglossus semilaevis) (csPGRP-L), a marine teleost with great economic value, was constructed through the comparative modeling method, and the key amino acids involved in the interaction with Lys-type PGNs and Dap-type PGNs were analyzed by molecular dynamics and molecular docking methods. Results: csPGRP-L possessed a typical PGRP structure, consisting of five ß-sheets and four α-helices. Molecular docking showed that the van der Waals forces had a slightly larger contribution than Coulombic interaction in the csPGRP-L-PGN complex. Moreover, the binding energies of csPGRP-L-PGNs computed by MM-PBSA method revealed that csPGRP-L might selectively bind both types of MTP-PGNs and MPP-PGNs. In addition, the binding energy of each residue of csPGRP-L was also calculated, revealing that the residues involved in the interaction with Lys-type PGNs were different from that with Dap-type PGNs. Conclusions: The 3-D structure of csPGRP-L possessed typical PGRP structure and might selectively bind both types of MTP- and MPP-PGNs, which provided useful insights to understanding the functions of fish PGRPs.

This work aimed to evaluate the role of the spleen in storage and reactivation of the memory B cells, represented by IgM positive cells and the systemic production of sheep antibodies anti-red cell in Nile tilapia (Oreochromis niloticus). Two groups were established: the memory group, containing fish previously immunized with a 2,5% sheep anti-red cell, to generate the immune memory; and the naive group, containing fish that received a 0,65% saline solution. After 32 days, both groups were subjected to a new dose of the same antigen at the same concentration, volume, and inoculation via. The memory clones reactivation was correlated to the increase of the IgM positive cells in the spleen in the memory group at 0 day. The memory group showed an increase in the absolute number of lymphocytes at 21 days and an increase in the antibodies at 14 days after inoculation when compared to the naive group. The results suggest that the spleen may be a storage and reactivation place of memory B cells in Nile tilapia.(AU)

RESUMO

In this study, the mobilomes of nine teleost species were annotated by bioinformatics methods. Both of the mobilome size and constitute displayed a significant difference in 9 species of teleost fishes. The species of mobilome content ranking from high to low were zebrafish, medaka, tilapia, coelacanth, platyfish, cod, stickleback, tetradon and fugu. Mobilome content and genome size were positively correlated. The DNA transposons displayed higher diversity and larger variation in teleost (0.50% to 38.37%), was a major determinant of differences in teleost mobilomes, and hAT and Tc/Mariner superfamily were the major DNA transposons in teleost. RNA transposons also exhibited high diversity in teleost, LINE transposons accounted for 0.53% to 5.75% teleost genomic sequences, and 14 superfamilies were detected. L1, L2, RTE and Rex retrotransposons obtained significant amplification. While LTR displayed low amplification in most teleost with less than 2% of genome coverages, except in zebrafish and stickleback, where LTR reachs 5.58% and 2.51% of genome coverages respectively. And 6 LTR superfamilies (Copia, DIRS, ERV, Gypsy, Ngaro and Pao) were detected in the teleost, and Gypsy exhibits obvious amplication among them. While the SINE represents the weakest ampification types in teleost, only within zebrafish and coelacanth, it represents 3.28% and 5.64% of genome coverages, in the other 7 teleost, it occupies less than 1% of genomes, and tRNA, 5S and MIR families of SINE have a certain degree of amplification in some teleosts. This study shows that the teleost display high diversity and large variation of mobilome, there is a strong correlation with the size variations of genomes and mobilome contents in teleost, mobilome is an important factor in determining the teleost genome size.

Abstract: The study of feeding habits in fishes can give important information about their role in the fish community, their functional role in the ecosystem, and their relationships with all available resources. We studied H. sierraensis from Gaira River and performed eight bi-monthly samples between May 2014 and November 2015. We analyzed fish stomach contents by the numerical (% N), volume (% V) and frequency of occurrence (% FO) methods; we also calculated the index of food importance (IA), coefficient of emptiness (CV), trophic niche breadth (used H') and condition factor (K). Additionally, we determined the sex ratio, gonadosomatic index (RGS), fecundity, minimum and average size at sexual maturity, and oocyte diameter by standard methods. The Gaira River has irregular meanders with clear waters, and high current velocity through its drainage; it is generally oligotrophic, with a tendency to eutrophic condition in some places. Observed dissolved oxygen values were high (5.4 mg/L), electrical conductivity was 68.63 µS/cm, and pH was found neutral. We analyzed stomach contents of 477 specimens, and found a 1.59 % coefficient of vacuity. The species was found euryphagic in its feeding habits (H'= 0.69), omnivorous tending to insectivorous; its diet consisted mainly of Trichoptera larvae (Atanatolica sp.), followed in importance by snails (Gastropoda Pomacea sp.) and dipteran larvae such as Simulium sp. Significant differences in both frequency of occurrence and the volume consumed were observed for the different size classes, as well as between different seasons of the year. The condition factor indicated that the species was well fed. Sex was determined for 424 fish, of which 241 were male and 183 female, given a ratio of 1.3: 1 (male-female) which resulted significantly different (Chi 2: 16.14; p= 0.02). The gonadosomatic index indicated that reproduction occurs during the high water river phase, fecundity was high, and oocyte diameter was relatively small. Males reached sexual maturity at an average size of 67.08 mm SL, and females at 70.68 mm SL. Their life history strategy is seasonal r. Management and conservation plans for this species are required due to human intervention in their habitat, originated by the growing tourist activity in the area. Rev. Biol. Trop. 65 (3): 1033-1045.

The retina of teleost fish zebrafish, has become an important model for studying neuronal plasticity and neurogenesis. It was further shown that the retina undergoes ontogenetic changes to adapt to different environments during their lifetime. This study aims to describe the ontogenetic development of the retina of juvenile salmon from hatching to the juvenile stage. We worked with 30 salmon divided into three groups of 10. Group I: newly hatched with yolk sac and 18 mm in length. Group II: without yolk sac and 30 mm in length. Group III: 100 mm long. Five fry each group were processed according to the protocol of Hanken & Wassersug to measure dorsoventral and nasal-temporal diameters using the cartilage that protects the eyeball. The remaining five specimens were sectioned with a microtome Microm serially (5 µm) and processed with technical H-E / Alcian blue. The layers of the retina were measured on a Zeiss optical microscope with camera Powershot built and with Image Tool 3.0 software. Group 1 showed large pigmented eyes, looking embryonic optic cup, the retina is stratified in layers. The inner nuclear layer (CNI) measured 62±10 microns and the inner plexiform layer (CPI) 10±2 µm. Group 2 presents changes in the thickness of them. The CNI decreases in thickness to 45±8 µm and the plexiform increased to 25±5 µm. In juvenile fish of group 3, the CNI reaches the minimum thickness (15±3 µm), by contrast, the inner plexiform layer thickness increases up to (70±10 µm). In the three groups observed in the periphery of the retina one proliferative germinative zone, which corresponds to a remnant of the embryonic neural epithelium responsible for the continued growth of the retina. The retina of the salmon can also be an important model for the study of ontogeny, neuronal plasticity and neurogenesis. This retinal neurogenesis fish rearrangement facilitates cell along ontogeny, potentially allowing optimization of the visual system to changes in the visual demands. This study may be useful to help diagnose pathologies in eye salmon and can also contribute to better understand tissue regeneration. On the other hand, with later studies, fish's retinal neurogenesis could be extrapolated to the treatment of human retinal diseases, such us glaucoma, retinal detachment o diabetic retinopathy.

RESUMO

Environmental changes affect fish homeostasis, turning them more susceptible to diseases. In Brazil, outbreaks of Streptococcus agalactiae infection have been reported in Nile tilapia when they are outside of their thermal comfort zone. This investigation evaluated mortality rate and which were the most infected organs at temperatures that naturally occurred in southern of Brazil. Forty Nile tilapia (Oreochromis niloticus) were infected with S. agalactiae and distributed in four groups (n=10) and each group was exposed to a different temperature: G1: 24°C, G2: 26°C, G3: 28°C, and G4: 32°C. Fish were monitored for 10 days. In this period, fish that presented irreversible clinical signs were sacrificed and samples of brain, liver, and kidney were collected for bacteriological and molecular analysis. Signs compatible with a streptococcal infection were observed in all groups. Highest mortality rates occurred at 24°C and 32°C. The brain was the most affected organ with the highest percentage of isolation of S. agalactiae by both methods of diagnosis. The results suggest that, as in mammals, temperatures that are further away from the comfort zone influence fish homeostasis, increasing susceptibility to bacterial infections.

In this study, histological, ultrastructural, and immunohistochemical observations of the gill tissue of two fish species (Plagioscion squamosissimus and Lithodoras dorsalis) were used as biomarkers to assess the ecological conditions of an estuary in the Amazon delta, by relating the health status of the fishes to environmental quality. The gill tissue was analyzed both qualitatively and semi-quantitatively. Marked differences in the type and severity of lesions were verified between species and among study sites. A variety of severe damages were confirmed histologically in specimens from the two impacted areas (B and C), including aneurysms, epithelial lifting, and intense hyperplasia. Electron microscopy and immunohistochemical analyses also revealed injuries in the gill tissue of specimens from these two areas. While, on area A only few specimens showed mild changes in gill tissues and rare marking of apoptosis. These results confirm the urgent need for the more effective control of the discharge of pollutants into the estuary as well as more effective urban planning, given the close proximity of a major industrial complex to residential areas.

The aim of this study was to characterize the normal structure of the liver of Hemisorubim platyrhynchos, a carnivorous freshwater catfish found in Neotropical region, using gross anatomy, histology and histochemistry. Anatomically, the liver presents C-shaped and only two lobes: smaller right and bigger left. The gallbladder is located in right lobe and shows elongated shaped. Histological analysis demonstrated that the hepatic parenchyma is made of two hepatocytes plates surrounded by sinusoids. The hepatocytes are polygonal-shaped cells, with spherical nucleus and a dark prominent nucleolus. The cytoplasm presents large amount of lipids and glycogen deposits PAS positives. There are no hepatic lobules or portal triads. Bile ducts are lined by columnar epithelial cells with apical mucosubstances PAS and AB positive. Furthermore, the liver presents melano-macrophages centers, distributed next to the blood vessels and bile ducts, constituted by cells accumulating pigments, whose presence may be related to the nutritional status of the fish. Moreover pancreatic tissue was observed in visceral portion of liver, formed by exocrine pancreatic tissue and islet organ, constituting an extrahepatic pancreas.

Objetive. This study was conducted to evaluate, by means of lectinhistochemistry (LHC), the expression of carbohydrates in granulomas induced by the bacillus Calmette-Guerin (BCG) in muscle tissue of Piaractus mesopotamicus after 33 days. Material and methods. Histological sections with 3 μm thick were incubated with the following lectins :WGA (Wheat germ agglutinin), DBA (Dolichos biflorus agglutinin) and HPA (Helix pomatia agglutinin), and the results were evaluated by light microscopy. Results. Acid fast bacilli were stained by Ziehl Neelsen (ZN) and strong labeled by WGA in the cytoplasm of macrophages. Labeling with DBA was intense in fibroblasts and weak in macrophages. On the other hand, HPA binding was stronger in macrophages, especially in those that were in close contact with epithelioid cells, without evidence of binding to fibroblasts. The epithelioid cells were not labeled by the used lectins, but they were identified by Hematoxilin-Eosin (HE). The lectins labeled specific type saccharides in glycoproteins, as N-acetylglucosamine present in bacilli and macrophages, as well as N-acetyl-galactosamine in macrophages. The control group showed no inflammation or lectin binding. Conclusions. This technique may be useful in identifying receptors for WGA, DBA and the HPA lectins in epithelioid granuloma induced by BCG in P. mesopotamicus.

Natural insecticides from plant extracts represent an alternative to the highly toxic synthetic products in order to reduce environmental contamination; however some might also be toxic for non-target organisms. The present study determined the 50% lethal concentration (48h; LC50) for adults Nile tilapia (Oreochromis niloticus) exposed to the natural insecticide Fumydro®, based on the tobacco plant (Nicotiana tabacum), and evaluated its effect on hematological variables. After preliminary tests, adult specimens of O. niloticus were exposed to four Fumydro® concentrations (200, 300, 400 and 500 µL L-1). The 48h; LC50 of Fumydro® was determined at 370 ± 50 µL L-1. The surviving fish after exposure to Fumydro® showed an increase in the number of red blood cells, hemoglobin concentration, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration. The number of thrombocytes and leukocytes has not changed, unlike the differential leukocyte count that presented an increased percentage of neutrophils. The results indicated that the insecticide Fumydro® is highly toxic to Nile tilapia and changes in erythrocyte variables suggested the induction to hypoxemia with low effect on the immune system.

A new ceratomyxid parasite was examined for taxonomic identification, upon being found infecting the gall bladder of Hemiodus microlepis (Teleostei: Hemiodontidae), a freshwater teleost collected from the Amazon River, Brazil. Light and transmission electron microscopy revealed elongated crescent-shaped spores constituted by two asymmetrical shell valves united along a straight sutural line, each possessing a lateral projection. The spores body measured 5.2 ± 0.4 µm (n = 25) in length and 35.5 ± 0.9 µm (n = 25) in total thickness. The lateral projections were asymmetric, one measuring 18.1 ± 0.5 µm (n = 25) in thickness and the other measuring 17.5 ± 0.5 µm (n = 25) in thickness. Two equal-sized subspherical polar capsules measuring 2.2 ± 0.3 µm in diameter were located at the same level, each possessing a polar filament with 5-6 coils. The sporoplasm was binucleate. Considering the morphometric data analyzed from the microscopic observations, as well as the host species and its geographical location, this paper describes a new myxosporean species, herein named Ceratomyxa microlepis sp. nov.; therefore representing the first description of a freshwater ceratomyxid from the South American region.

RESUMO

The use of aluminum silicates for decontaminating animal feed containing aflatoxins has yielded encouraging results in chicken and turkey poults. In contrast, very few studies have tested these substances in aquaculture. In this work, we investigated the efficacy of a trout diet containing 0.5 percent hydrated sodium aluminosilicate (HSAS) in protecting against contamination with aflatoxin B1. Trout were reared on these diets for one year and the experimental groups were examined monthly for hepatic presumptive preneoplastic and neoplastic lesions. Regardless of the presence of HSAS, all of the fish that received aflatoxin in their diet have shown hepatic lesions indicative of a carcinogenic process, presenting also the development of cancer in some fish. The concentration of HSAS used in this study was ineffective in preventing the onset of hepatic lesions induced by aflatoxin B1 in rainbow trout.
